HER2調(diào)研報(bào)告-12017/12/24HER2BackgroundHER2SignalingPathwayHER2Market蛋白開品開發(fā)戰(zhàn)略產(chǎn)品推廣方案ErbB2,alsocalledNeuandHer2(humanepidermalgrowthfactorreceptor2),isatypeImembraneglycoproteinthatisamemberoftheErbBfamilyoftyrosinekinasereceptors.ErbBfamilymembersserveasreceptorsfortheepidermalgrowthfactor(EGF)familyofgrowthfactors.ErbB2iswidelyexpressedinepithelialcellsandhasalsobeenfoundtobeover-expressedinalargenumberofbreastcarcinomas.AmongErbBfamilymembers,ErbB2isuniqueinthatithasnoidentifiedligands.Rather,ErbB2heterodimerizeswiththeothermembersoftheErbBfamily(ErbB1(EGFR),ErbB3,ErbB4)toformhigheraffinitysignalingcomplexes.BecauseErbB3containsadefectivekinasedomain,thekinasedomainofErbB2isresponsibleforinitiatingthetyrosinephosphorylationsignalthroughtheheterodimericreceptor.IthasbeenfoundthatadiscretethreeaminoacidsignalintheErbB3cytoplasmicdomainiscriticalfortransactivationofErbB2.Interestingly,thissamethreeaminoacidsignalhasalsobeenfoundinErbB1andErbB4.Phosphoinositide3-kinasehasbeenshowntoplayaroleinErbB2signaltransduction.ThecytoplasmicdomainofErbB2hasbeenshowntoassociatewithbeta-cateninandplakoglobin.HumanErbB2consistsof1255aminoacids(aa)witha21aasignalsequence,a631aaextracellulardomain,a23aatransmembraneregion,anda580aacytoplasmicdomain.ErbB2canbeshedfromthecellsurfacebyproteolyticcleavagebyanunidentifiedprotease.ErbB2appearstoplayrolesindevelopment,cancer,communicationattheneuromuscularjunctionandregulationofcellgrowthanddifferentiation(1-10).HER2BackgroundHER2SignalingPathwayEGFR/ErbB1ErbB2/Her2ErbB3/Her3ErbB4/Her4FGFR1FGFR1-4FGFR1alphaFGFR1betaFGFR2FGFR2alphaFGFR2betaFGFR3FGFR4HGFR/c-METIGF-IRIGF-IIRInsulinandInsulin-likeReceptorInhibitorsInsulinR/CD220INSRRPDGFRalphaPDGFRbetaTie-1Tie-2TrkATrkBTrkCVEGFR1,R2,R3VEGFR1/Flt-1VEGFR2/KDR/Flk-1VEGFR3/Flt-4HER2Market截止到20171218RegionName
