1、普洱茶茶褐素降血脂功效及其作用機制研究摘要 本實驗探討了普洱茶茶褐素對高脂SD大鼠的降血脂作用及其作用機理,采用生化試劑盒研究普洱茶茶褐素對實驗大鼠血清中TC、TG、HDL-C、LDL-C、LCAT、LPL、HL的動態(tài)影響,檢測了普洱茶茶褐素對肝臟和附睪組織中HSL活性及其mRNA表達量的影響,分析了普洱茶茶褐素對高脂飲食大鼠糞便中膽固醇和膽汁酸含量的影響,通過電子顯微鏡觀察了普洱茶茶褐素對實驗大鼠肝臟病理變化的影響。結(jié)果顯示:1、經(jīng)飼喂高脂飲食4周后,模型組大鼠血清TC、LDL-C顯著升高(P0.01)、TG顯著升高(P0.05),說明本實驗所采用的高脂血癥大鼠模型是成功的。2、與高脂模型組
2、相比,治療組高脂血癥大鼠血清TC、LDL-C水平顯著降低(P0.01),TG顯著降低(P0.05)、HDL-C水平明顯升高。說明普洱茶茶褐素具有降血脂作用;預防組高脂飲食大鼠血清中TC、TG、LDL-C水平升高HDL-C水平降低受到抑制,與空白對照組沒有顯著性差異(P0.05),說明茶褐素能減少肝臟脂肪沉積,具有預防實驗動物脂肪肝形成的作用;正常試物組大鼠血清TC、TG、HDL-C、LDL-C水平與空白對照組相比也沒有顯著性差異(P0.05),這說明茶褐素對正常大鼠血脂水平影響不顯著。3、普洱茶茶褐素可以增強高脂血癥大鼠HL、HSL活性,其中HL活性上升了62.50%,肝組織和附睪組織中分別上
3、升了51.00%和163.41%。同時,茶褐素能顯著提高大鼠肝組織和附睪脂肪組織中HSL的mRNA表達量,表明茶褐素能通過調(diào)節(jié)脂質(zhì)代謝關(guān)鍵酶活性來發(fā)揮其降脂功效,基本明確了茶褐素降脂作用的酶學和分子生物學作用機制。4、測定結(jié)果表明,茶褐素促使糞便中膽固醇和膽汁酸的含量分別增加了21.11倍和4.08倍,與高脂模型組相比,差異極顯著(P0.01)。說明普洱茶茶褐素具有抑制大鼠體內(nèi)外源性膽固醇的吸收,加速其排泄的作用。5、對不同發(fā)酵階段茶褐素的降血脂效果進行了研究,結(jié)果表明四翻茶褐素的降血脂效果較好。關(guān)鍵詞:普洱茶;茶褐素;降血脂;作用機制Study on the hypolipidemic fu

5、etect dynamics of lipid contents in serum.HSL mRNA expression and HSL activity were used to detect in liver and the fat of epididymis.The cholesterol of the stool,fecal bile acids were measured. The results include:1、It shows in this subject that the amount of TC in the blood serum of the rat fed on
6、 high fat diet for 4 weeks and the amount of TC,LDL-C in the blood serum increase remarkably(P0.01),the amount of TG in the blood serum increase remarkably (P0.05),which indicates that the rat served as hyperlipdemia animal model are successful.2、As compared with high fat control group, the amount o
7、f TC, LDL-C in the blood serum of the treats rat decreased remarkably(P0.01)and the amount of TG in the blood serum decreased remarkably(P0.05),the amount of HDL-C in the blood serum decreased. The result is that TB can decrease the amount of lipid;This study manifested that TB can prevent the amoun
8、t of TC、TG、LDL-C from the increase,moreover preventing the amount of HDL-C from the decrease in blood lipid of high-fat-diet rat.In contrast with normal control group, there are not significantly different(P0.05).It shows that TB can reduce sediment of fat in liver and protect the form fatty liver;I
9、n contrast with normal control group,TC、TG、HDL-C 、LDL-C levels in nomal test group are not significantly diefrent(P0.05).It shows that TB has not effect on blood lipids levels of normal dietary rat.3、The HL and HSL activity were significantly increased by TB feeding. The activities of HL and HSL in
10、liver and the fat of epididymis were increased by TB(62.50%、51.00%、163.41%).The mRNA expression of HSL were significantly increased in liver and the fat of epididymis.It shows that TB can activate the activities of lipid metabolism to decrease the amount of lipid.The mechanism of Enzymology and Mole
11、cular Biology were studied on hypolipidemic function of TB.4、It shows that the cholesterol of the stool,fecal bile acids were significantly increased by 21.11 times and 4.08 times. In contrast with high fat control group, there are significantly different (P0.01).
