27、2. 遼寧中醫(yī)藥大學(xué)教學(xué)實(shí)驗(yàn)中心,沈陽 110847)SET bkTitleInfo * MERGEFORMAT SET bkAuthorsInfo * MERGEFORMAT 摘要: FORMTEXT 目的:觀察補(bǔ)腎、健脾和補(bǔ)腎健脾三方對(duì)模擬失重大鼠腰椎骨的影響,比較其防治作用的異同。方法:大鼠隨機(jī)分為正常對(duì)照、懸吊、補(bǔ)腎、健脾和補(bǔ)腎健脾共5組。后4組大鼠頭低位-30度尾部懸吊連續(xù)21d,補(bǔ)腎、健脾和補(bǔ)腎健脾組大鼠從實(shí)驗(yàn)第1天開始依次給予補(bǔ)腎方、健脾方和補(bǔ)腎健脾方灌胃,其余各組大鼠灌服等容積的生理鹽水。實(shí)驗(yàn)第21天處殺各組大鼠,雙能X線骨密度儀測(cè)L4椎體骨密度,L4椎體麗春紅三色染色觀察骨形

28、態(tài)計(jì)量學(xué)靜態(tài)參數(shù)。結(jié)果:較之正常對(duì)照組,懸吊組大鼠L4椎體骨密度顯著降低(P0.01),骨小梁面積百分率、骨小梁厚度、骨小梁數(shù)量明顯降低(P0.01),骨小梁分離度明顯增高(P0.01);較之懸吊組,補(bǔ)腎組和補(bǔ)腎健脾組大鼠L4椎體骨密度顯著升高(P0.01),骨小梁面積百分率、骨小梁厚度、骨小梁數(shù)量明顯升高(P0.01),骨小梁分離度明顯降低(P0.01),健脾組大鼠L4椎體骨密度升高(P0.05),骨小梁面積百分率、骨小梁厚度明顯升高(P0.01),骨小梁數(shù)量增加、骨小梁分離度降低(P0.05);較之補(bǔ)腎健脾組,補(bǔ)腎組大鼠L4椎體骨密度降低、骨小梁厚度降低升高(P0.05),健脾組大鼠L4椎

32、action of tonifying kidney prescription, invigorating spleen prescription and tonifying kidney and invigorating spleen prescription on the lumbar vertebra in tail suspension model rats. Methods: Rats were assigned to five groups randomly: control(C),tail suspended(S),tonifying kidney (TK), invigorat

33、ing spleen(IS) and tonifying kidney and invigorating spleen (TKIS) groups. Rats in S,TK, IS and TKIS groups were suspended by head down tilt -30for 21 days. Rats of TK, IS and TKIS groups were orally administrated with three decoctions and other groups with equivalent saline from the first day. Bone

34、 mineral density of lumbar 4 was measured respectively using dual energy X ray absorptiometry in rats on day 21. Meanwhile bone histomorphometry static parameters of lumbar 4 in rats of C,S,TK, IS and TKIS groups were measured by ponceau trichrome staining. Results: Compared with the C group, bone m

35、ineral density, trabecular area percentage, trabecula thickness and trabecula number of L4 are decreased and trabecular separation of L4 is increased obviously in S group. Compared with the S group, the bone mineral density, trabecular area percentage, trabecula thickness and trabecula number of L4

36、are raised and trabecular separation of L4 is reduced in TK, IS and TKIS groups. Compared with the TKIS group, the bone mineral density, trabecular area percentage and trabecula thickness of L4 are decreased and trabecular separation of L4 is increased in TK and IS groups.Conclusion: Three prescript

37、ions including tonifying kidney prescription, invigorating spleen prescription and tonifying kidney and invigorating spleen prescription can protect the bone loss induced by simulated weightlessness and the role of tonifying kidney and invigorating spleen prescription is superior.Key words: FORMTEXT

38、 Formula-ology; tonifying kidney and invigorating spleen, simulated weightlessness, tail suspension,bone loss引言大量研究表明,航天員在進(jìn)入太空失重環(huán)境飛行過程中,骨質(zhì)丟失是一種必然發(fā)生的病理改變1、2。至今,尚未研究出對(duì)抗這種骨丟失安全有效的措施。近年來,祖國(guó)醫(yī)學(xué)在此方面多采用以補(bǔ)腎為主的方藥進(jìn)行干預(yù),取得了良好效果。然而中醫(yī)認(rèn)為“腎主骨,脾主四肢”、“脾主運(yùn)化,充養(yǎng)腎精,精生髓,髓養(yǎng)骨”。脾和腎對(duì)機(jī)體骨量維持恒定均具有重要作用,因此本研究在建立大鼠尾部懸吊模擬失重骨丟失模型的基礎(chǔ)上,

39、著重探討補(bǔ)腎、健脾和補(bǔ)腎健脾三方對(duì)模擬失重腰椎骨丟失的防護(hù)作用,比較其異同,力求獲得有效的防護(hù)方藥。材料與方法材料動(dòng)物健康雄性Wistar大鼠60只,級(jí)別SPF/VAF,體重(18010),北京維通利華實(shí)驗(yàn)動(dòng)物中心提供。許可證號(hào):SCXK(京)2006-0009。藥物方劑補(bǔ)腎方:由鹿茸、淫羊藿等組成,健脾方:由生曬參、生黃芪等組成,補(bǔ)腎健脾方:由生曬參、生黃芪、鹿茸、淫羊藿等組成,全部藥材購(gòu)自北京同仁堂遼寧有限責(zé)任公司。補(bǔ)腎方和補(bǔ)腎健脾方中鹿茸打碎,過100目篩兌入藥液,三個(gè)方中其余藥物均采用常規(guī)水煎煮法提取,補(bǔ)腎方、健脾方和補(bǔ)腎健脾方將所得全部藥液混合依次濃縮成0.42g原藥材/ml、0.5

40、6g原藥材/ml、1g原藥材/ml濃度備用。儀器雙能X線骨密度分析儀:美國(guó),Lunar;DMLA顯微照相系統(tǒng):德國(guó),Leica;硬組織切片機(jī):德國(guó),Leica。方法動(dòng)物分組與處理大鼠隨機(jī)分為5組:正常組(C)、懸吊組(S)、補(bǔ)腎組(TK)、健脾組(IS)和補(bǔ)腎健脾組(TKIS),每組12只。依據(jù)人和動(dòng)物間體表面積折算的等效劑量比值表計(jì)算大鼠給藥劑量,TK、IS、TKIS組大鼠從實(shí)驗(yàn)第1天開始依次按2.4 gkg-1d-1、3.2 gkg-1d-1、5.7 gkg-1d-1給予補(bǔ)腎方、健脾方和補(bǔ)腎健脾方灌胃。其余各組大鼠灌服等容積的生理鹽水。S組、TK組、IS組、TKIS組大鼠采用頭低位-30尾

41、部懸吊法3模擬失重,每天懸吊24h,共懸吊21d;C組大鼠不采取任何處理。實(shí)驗(yàn)第21天,各組大鼠稱重,于灌胃后2h脫頸椎處死,取第4腰椎。指標(biāo)測(cè)定腰椎重量測(cè)定除凈附在腰椎4上的肌肉和軟組織,在分析天平上稱量重量,計(jì)算大鼠L4椎體占體重比(mg/g)。骨密度測(cè)定每組中隨機(jī)選取6只大鼠的L4椎體,生理鹽水沖洗,用雙能X線骨密度分析儀,測(cè)定大鼠離體腰椎4的骨密度,以單位面積的骨礦物質(zhì)含量(g/cm2)表示骨密度值。骨形態(tài)計(jì)量學(xué)參數(shù)測(cè)定每組中剩余6只大鼠的L4椎體,用10%中性甲醛固定,70%-95%酒精梯度脫水,塑料液浸泡,塑料包埋,用硬組織切片機(jī)切片,厚度30m,麗春紅三色染色,用全自動(dòng)顯微鏡在5

42、0倍視野下進(jìn)行圖像采集,取切片中心部的五個(gè)視野測(cè)量并取其平均數(shù),Image Pro Plus6.0軟件測(cè)量骨小梁面積比(Tb.Ar)、骨小梁厚度(Tb.Th)、骨小梁數(shù)量(Tb.N)、骨小梁分離度(Tb.Sp)。統(tǒng)計(jì)學(xué)處理所得數(shù)據(jù)均用(s)表示,用SPSS 11.0 軟件處理,單因素方差分析(one-way ANOVA)后,行多組間比較,采用Student-Newman-Keuls檢驗(yàn)。結(jié)果補(bǔ)腎、健脾、補(bǔ)腎健脾三方對(duì)模擬失重大鼠L4椎體重量的作用結(jié)果見表1。與C組比較,S組大鼠L4椎體重/體重明顯降低(P0.01);與S組比較,TK、IS、TKIS組大鼠L4椎體重/體重均明顯升高(P0.01)

47、體骨組織的麗春紅三色染色(50)A:正常組B:懸吊組C:補(bǔ)腎組D:健脾組E:補(bǔ)腎健脾組討論實(shí)踐證明,人類在進(jìn)入太空失重環(huán)境后,會(huì)出現(xiàn)骨質(zhì)丟失,導(dǎo)致骨密度下降和骨質(zhì)疏松。太空飛行一個(gè)月導(dǎo)致骨密度下降約1-1.5%,這些骨質(zhì)丟失主要發(fā)生于某些特定部位如腰椎、股骨頸、股骨大轉(zhuǎn)子和骨盆,其程度較絕經(jīng)期骨質(zhì)疏松更為嚴(yán)重4。為了提高航天員飛行中的工作能力和返回地面后的再適應(yīng)能力,必須采取有效的防護(hù)措施。本研究所采用的大鼠尾部懸吊模型,是目前國(guó)內(nèi)外公認(rèn)的研究模擬失重常用的地面動(dòng)物模型5。實(shí)驗(yàn)結(jié)果顯示,大鼠尾部懸吊21天L4椎體骨密度顯著下降,骨形態(tài)計(jì)量學(xué)靜態(tài)參數(shù)改變:骨小梁面積百分率、骨小梁厚度、骨小梁數(shù)量明顯降低,骨小梁分離度明顯增高,L4椎體與體重之比也明顯下降,與以往的研究結(jié)果一致6,表明大鼠尾部懸吊21天出現(xiàn)了明顯的骨質(zhì)疏松。失重引起的骨丟失是古代中醫(yī)學(xué)從未涉及的病癥,是現(xiàn)代人在探索未知世界面臨的一項(xiàng)亟待解決的醫(yī)學(xué)課題。但是中醫(yī)學(xué)對(duì)“骨”的生理有相關(guān)記載:如“腎主骨,脾主四肢”、“脾主運(yùn)化,充養(yǎng)腎精,精生髓,髓養(yǎng)骨”。因此對(duì)失重性骨丟失可以
